I am using a Streptavidin agarose system to affinity purify biotin labeled, outer membrane proteins of a gram negative bacterium. Could you provide info regarding preparation of buffers and the reaction and elution conditions of this system?
The high affinity of streptavidin for biotin precludes using streptavidin-agarose for binding and eluting biotinylated proteins, because there is no way to elute the proteins without denaturing them and the streptavidin.
Streptavidin-agarose has been used for affinity purification, by binding a biotinylated affinity ligand. Examples include binding biotinylated antibodies to streptavidin-agarose for immunoaffinity purification of
